Selection of Affinity Tags for Recombinant Protein Purification

Abstract

The in vitro recombinant protein expression technology has penetrated all fields of biology. At present, four major types of in vitro recombinant protein expression systems are used: prokaryotic expression system, mammalian cell expression system, yeast expression system, and insect cell expression system. In general, the expression procedure includes three steps: vector construction, expression identification, and protein purification. During the vector construction stage, codon optimization and tag selection also need to be considered in addition to confirming the necessary expression elements. Hence, choosing a suitable affinity tag is not only conducive to the overall protein purification procedure but will enhance protein solubility while maintaining the structure and function of the protein for downstream applications. This paper introduces several protein purification tags in detail to help better design the experiment.
